(sub)domains are registered, and may not always be running on the same server. DNS zone files tell which IP addresses are mapped to the domain. If the domain is a wildcard, then you could use a script to parse through the apache configuration files to determine how many subdomains existed.

At this point, any domain that makes it's zone file available to the public should be considered broken. This has been the case for at least the last 10 years.

There is no way to tell using DNS, other than guessing.

Search (Google, etc.) could be used, but will only reveal subdomains that the search engine knows about. If there are no links in to the subdomain, the search engine won't (and can't) know about it.
