Submitting same domain with different docs or directories

May I? site/regional1/ or site/regional.htm?

Scenario:
Travel Site deals services in different cities.
The main product or service would be the guide itself,
but the most acurrate searches are the regions inside the site.

So I wonder if making that structure allows to submit every region as a separate site to search engines and directories (let's say dmoz in principle)

www.site.com > 1 submit
www.site.com/region1.htm > other submit
www.site.com/region2.htm > other submit and so on...

Or this one:

www.site.com > 1 submit
www.site.com/region1/index.htm > 1 submit
www.site.com/region2/index.htm > 1 submit and so on...

Is that SE friendly?

What about

www.site.com
region1.site.com
region2.site.com?

Finally: What about English and Foreign language versions?

Should I submit twice in order to take advantage of keywords and because many directories and SE only allow english sites?

www.site.com (Spanish)
www.site.com/region1documentinenglish/ (English)

silverbytes, we've had discussions that are very closely related, if not exactly about what you're looking at.

Do a site search for subdomain + directory (use plurals and singular) and you'll come up with a ton that'll take days of reading. You'll also want to check out "duplicate content" and cross-linking, which are both issues that have come up with regard to using subdomains - or separate domains vs. subdomains, for that matter.
