Is it best to separate keywords and keyword phrases by commas, or not? Should keywords be placed in the Title tag?

I think that it is safe to say that you want to put your keywords or kw phrae in your title tag. I'm not quite sure what you mean by using commas.

Its important to have the key words and key phrases in the title tag to have a better rank in SE results.

Regarding use of comma:

Write the title in such a way that user gets to know in brief what the site is about.

It doesn't matter whether or not you use comma.
