It I buy a website today then can I add one oneway backlink everyday?
or one link a day is it too much for a new website?

I'd say almost any regular pattern, over time, will appear unnatural to the algorithms that Google uses. If you the owner can control the addition of a backlink, then it's not the kind of backlink that Google wants to value with a lot of strength.

And you are so lucky to have 50 backlinks you can add whenever you want.
When will you add them (in the fastest way possible)?

5 to 9 links a day, targetting different areas (blogs,forums,comments,directory, articles,social-networking, etc.)

I only add a few links per month, but this is because I don't know of a way to add more. My websites are doing OK though and links I add are most of the time from quality websites.

The pattern of link addition should be irregular which can be seen as natural. Also,this does not mean addition of 100 links in one day and none on the other.

Your site might get to the top of digg, attract say 10k visitors in one day and receive 10 links from some popular blogs in your niche. Then some 30 second tier blog in your niche catch up and you get another 30 links. Your site gets to forums, facebooks, etc. and you get some more backlinks during the next 10-15 days.
Possible? Possible.
Natural? Perfectly natural.

So now the key is to decide whether the 50 links you have are some crap links on pay-per-post blog with page rank or some really huge link opprotunities.

my backlinks are mostly from forums which I noticed have a nofollow tag. Does that help in any way other than the very few referrals?

My backlinks are oneway dofollow comments on blogs with good pr and to balance such good pr I add my websites to pr0 directories that do not have pr5 or pr6.
But I'd like to speed up such procedure so I was wonder what was the fastest way to submit such 50 oneway backlinks.
