This new version has no security fixes, so it's not necessarily a critical upgrade. There is a security enhancement of sort, in that 2.6 is designed to be fully compatible with SSL if you need that.
Other features:
- post from anywhere
- wiki-style revision tracking.
- google gears
- image captioning
- 194 bug fixes.
For me, I have to say, nothing compelling, but the 2.5 branch will not be maintained, so if you want to keep up on security and bug fixes, you'll need to go to 2.6.
WP says that it should be fully compatible with 2.5 plugins and themes, but Joost de Valk for one had to make several bug fixes to his plugins that came to light as a result of the upgrade.

I'll be working on upgrading a client's site to the newest version of WP in a week or so. Was planning on WP 2.5 but with this new release I'm reconsidering that we might want to just upgrade to 2.6 if we have to upgrade anyway. I'm just concerned about the plug-ins being an issue as they usually are of concern when upgrading. I hope everything goes smoothly with this new version, otherwise, I'm going to have to resort to just 2.5, which isn't that bad, anyway.
