Can I have two identical ad groups; same ad title, same ad text, same display URL, same keywords, but different destination URLs in one campaign?

Yes, I do it to "load balance" users to two separate AsiaPac mirrors of the same site.

It CAN work this way, but that's not the optimal solution. Google doesn't "like" it....
You better create two identical ads in the same group, each one can have a different landing page.
