My site is primarily US and hosted in the US but is a general topic so it doesn't have a strict connection with any country. However, to lessen the low paying countries would it be better if I set targeting to US in webmaster tools?

Probably wouldn't help, particularly with interest based ads. People from other countries are going to see ads targeted to them. The webmaster tools target is for organic traffic. Now, if you mostly only want US traffic, then you could set it, but I'd be surprised if you saw a revenue change after you did.
