Do I have to generate the ad code every time I want to change the colours?

If I want just to change colors on my ads, do I have to re-generate them again on the adsense setup page, or can I just put the new hex values by hand?

I mean, does it matter at all?

Probably won't make a difference just for the colors, but:
In order to display Google ads on your website, it is very important that you copy the ad code exactly as it appears on the AdSense Setup tab of your account and paste it directly into the HTML source code of your webpages, without any modifications. Modifying the ad code is against our program policies and may also result in errors on your page.

Of course.
The Adsense code is only a string of alphanumeric characters.

Just don't make a mistake, even to the extent of including a hash mark ('#') when specifying the colour code.
