I have just started using Adsense on a directory. Without going in to specifics, click-through rates are reasonable compared to other sites I have used it on.
The ads are usually on topic. I use the 468x60 banner so there are only two ads and sometimes just one. The directory has 20 links per page with a short description, which should give you some idea of the amount of content.
The directory is only around 120 pages or so at the moment. Hopefully the Adsense revenue will grow as the site does, although I have a banner ad server in place to eventually target advertisers directly and possibly not use Adsense at all.
mediabot does not pick any of the text from the title or description of directory listing and one of the directories does not have much text on the pages and it did take a little time to change the psa ads to ads relevent to the pages.

Google know what sites I have the ads on so I can only assume the answer is yes
when you open an account they check your site, then you can use the same code on as many sites as you require because they track the ads via mediabot
I had no problems getting the first one listed and just added to code to the others, not had an email from google so everything must be OK

This is in marked difference to the other directory which has about 73K pages (with another 50K or so that have not been published yet) and has Adsense running on approximately 36K of these pages. It has an ODP subdirectory of Irish/Irish related websites but I have not added Adsense to that because I have yet to go through it for quality control/404 checking.
The problem with any directory is the traffic though. It does not matter how specialised the directory is when there is no traffic.
I've mainly used skyscraper for both sites for the last few months. However I have switched about 27K pages to leaderboard on the big directory over the past few days.
