I realize mobile rankings are penalized, Are the DESKTOP search ranking penalized with mobile unfriendly sites?

Put simply, no.
In fact, desktop rankings practically dictate where mobile-friendly rankings would appear, so desktop rankings are more important than mobile as far as the current algorithm goes. There may well be less impact on mobile rankings that you might expect, as the PR has not really matched the reality when it comes to Google's handling of mobile sites.
That said, you're likely looking at 50% of your search audience being mobile, so best to sort it out ASAP ;)
