I want to change the title, description pertaining to keywords.
Now my question is if I change the title, description for all the pages will page rank get affected.

I often wondered about that. Do you know that for a fact? Does that mean that the more that link to you, the higher your PR will be?

Today I observed my main site page reduced in ranking by 1. And all the topic specific pages reduced their PR from 6 to zero!
What is happening here? Any clues...

Well this is mostly true, but keep in mind that pr can be effected by page factor - i.e. filters. For example if you have a spammy site packed full of keywords and perhaps duplicate content - then backlinks will not be the only factor in pr - in this case the pr will most probably be reduced due to the page factors!
Correct me if this is not the case.
In my opinion - yes, title tags are very important (but most probably will not effect pr directly) - hence the bold on them in search results! and even meta description is sometimes used. Keywords probably not so important on most search engines these days though.
Being all my pages with PR2, my site is nowhere in SERPs.
Can you please clarify another doubt, changing the title, Meta desc and Meta kw as per seo rules, increase my SERPs or should I concentrate only on building good backlinks and wait for bots to crawl my website.

Actual PageRank is updated more frequently in the background - toolbar just takes more time to catch up and reflect the changes.
The only way on page optimisation can affect your PageRank is if some of the sites who link to you get penalised, causing a reduction in the inbound PR to your site.
Your page ranking (ie it's positioning in the results pages) is affected by many factors including PageRank (PR), title tags, etc etc etc.

Yes. It's not a set figure like 10 sites = PR1, 20 sites = PR2, but that's the basic concept; the more sites that link to you, the higher your PR will be.
Also keep in mind that the PR of a page linking to you affects how much PR your page receives from it. So 10 PR5 links are better than 10 PR1 links.

Could be a number of things. Possible penalty. Possible URL changes at your end.
just the pattern (and possibly history) of links to the site
While pattern and history of links can and most likely are factors in ranking, I don't think they affect PageRank.

If it's a new site, then it could be sandboxed, which is a whole other issue.

Think you have misunderstood something. SERPs = Search Engine Results Pages - you don't have a lot of control over those! ;)
Build quality (non spammy) backlinks at a consistant rate (ie not all at once), add new content - don't tinker with your site until it's ranking for key terms.

Yes. Inbound links are the only thing that impact PR (directly).
I think some people here are confusing PageRank (PR - the little green bar) with page ranking (your position in the SERPs for a keyword) - the two are not the same.
