You need to get the eye focus to the ad. So it depends on your website layout. There is an interesting eyetracking research discussed at [webmasterworld.com...]

In my website layout it is easier to integrate the rectangle format close to my text where the user is focussed at, so 300x250 is better in my situation.

I think it's ok to give this as it is a part of the discussion and has nothing to do with my site.

They definately seem to think the more it blends with your site the better.

I've tried both size rectangles and frankly I haven't seen a difference. I've gone to the medium one because it saves a little space for people with small screens.

medium rectangle works insanely better for me than the sky banner.

It all depends on your site and your content layout. I use both. I have the rectangle (300x250) doing much better than the banner-sizes on one site, while I have the banners doing better than the rectangles on another site. You just have to experiment, and use channels to track the results. Once you figure out a good combination, stick with it. You can drive yourself crazy trying to tweak it every day. Instead, once you get a decent CTR, focus on developing more content and bringing in more users...
