If I write in Adgroup negative keywords a certain expression, such as: "New York hotels," will it cover all the options, or do I need to include "hotels new york" as well?

You could use the negative for -new which would also filter out "new hotels", "new jersey hotels", "new england hotels" or all combinations including "new york".
You could use also use -york which would filter out"new york hotels" or "hotels in new york".
