I am creating a directory that I am hoping it will become large. What is the best way to structure the site. Should I make a basic HTML site using our GoLive software? I don't know any of the database programs but is this a better way?

No, you will need a script to run it efficiently. You will have to plan well and start out with a script that can/will grow with you allowing easy admin of all tasks involved. Here are a couple past threads for you to read that may be helpful:
