I was just wondering some of your thoughts on what time frame you use to gauge the effectiveness of changes you have made to your ad layous.

At least a week, unless you are confident you can pick up small changes in the middle of random daily variation.
I think it depends partly on your site. On high-traffic sites, especially with at least a few month's of history to compare to, you might need less time.
