How to sell ads direct

without impacting AdSense

I've just been given an opportunity to sell ads direct for a short period of time, that is for an advertiser's very specific ad campaign. I'm running Adsense ads on the main section of my website, and those pages are all dynamically generated (my own custom CMS). Basically it is one template for many pages. I am using AdManager on a secondary portion of my site, so I could implement the AdManager code on the main portion. But my concern is that if I make this switch for a month or so that I will significantly disrupt my AdSense account/earnings. Also, I have only ever used AdSense to fill my AdManager inventory, are there any recommended resources or tutorial to show how to manage direct ad sales through AdManager.

When I sell a placement, I switch from native Adsense code to Ad Manager with Adsense fallback, and set the direct campaign as 'Sponsorship.' If there is enough demand for your website, taking a placement out for a while doesn't affect much, and sometimes it pushes earnings for the rest of placements higher.

Do you switch to AdManager code for the entire website during the campaign, or just a portion of it?

You should make your life easier by switching completely to GAM and serve AdSense via GAM's dynamic allocation. Either by enabling it for ad units or by making AdSense line items.
