Has anyone switched from the large 336x280 rectangle to the medium 300x250 rectangle...
I know you asked this about image ads, but I wanted to chime in that when I tested 336X280 vs. 300X250 text ads, the latter won. I got higher CTR and EPC. It wasn't a huge difference, but enough that when I made the change on hundreds of pages, I increased my earnings by about $10/day.
Test, test, test! Can't be said enough. Find out what combination works best for YOUR particular site(s).
This means that if the person that visits your site clicks the top ad on the top AdSense block, then you will get paid more than if they click the bottom ad of the bottom block. The advertiser has bidded higher to be placed in a more prominent position. Actually, you'll get paid more than the bottom ad of the same block as well.
At least, that's how it works with the search results, so I asssume that it is the same with content AdSense.
All of the above suggestions are good ideas. You might also want to use custom channels to track the performance of different ad formats, colors and placements throughout your site. In this way, you can learn which ads your users are most likely to click, and apply this knowledge throughout your site.
