Yes, in AdWords, if you turn on auto tagging, you can use your straight URLs (and let AdWords auto append the required tagging data).

Outside of AdWords, for other traffic sources (email, Bing Ads, Facebook, etc) you'll have to manually append the utm parameters to your links.
