The most important thing you can do with any site is create really good content. Its the content that gives users a reason to visit. When you have great content you can then attract good quality inbound links.

Regarding Twitter and getting more followers, What you can do is use the twitter search to find other users who are tweeting about your theme/topic. Follow them and hope they follow you back.

Don't waste time Tweeting to much, If you fill a users time line they are likely to unfollow you. Only Tweet something you think will be well received, and no more than a few times a day.
