Long term effectiveness comes from lots of links, many of them not even showing on a links: search because maybe they are PR2 or PR3. But if you have a lot of these little links, you're not likely to lose big and fast either. Also, the PR algorithm doesn't look at "sites", it looks at pages. Your own site also has pages with links, and creating more content pages on your own site can also boost the PR of other pages on your site - as well as giving you more link text to work with. Not every valuable link comes from another domain - some can come from your own. And more content pages on a site gives you more chances to rank on more searches.
The thread asks two questions - 1) most effective and 2) fastest way to increase PR. As others here said, the fastest is probably get a few inbound links from high PR pages. But is that really effective? It's putting all your eggs in just a couple of baskets, and highly visible ones at that.
