Is it possible to utilize google search/google custom search/ google adsense search somehow to search withing a directory, eg under domain.com/mydirectory/*
So every url under /mydirectory/ is included in the search?

Yes - if you're setting up a custom search engine, using what Google Custom Search Help calls "URL patterns" should do this for you.

Just do a site search of the desired directory and add your search terms afterwards.
If you want to search, say, for "hilltop", within the Google Search sub-directory on WebmasterWorld, try this search [google.com]...
