make sure the jumpers on the drive are set to the correct position and your OS is running in plug and play mode...

If there's no DOS/Windows partition on the drive, it won't have a drive letter.

If you use win95 or 98 then you need to use a DOS utility called fdisk to create partitions. If you use windows xp then you can go to:
Control Panel -> Administrative Tools -> Computer Managment -> Storage -> Disk Managment and create a parition there.
