Help with sub-domain server setup

Two questions, should each sub-domain have a root directory; and if so how should this be setup on the server? Any other tips on setting-up sub-domains appreciated.

If that doesn't do it I'll have my host provider come back in and tell you how she's doing it because I really love how it works.

Thanks Paynt, I think that second thread might do the trick. As far as i know my host will supply individual IPs for subs.
